Histone acetylation is a key process in chromatin remodeling during development, but it is unclear how acetylation of histone N-terminal tails affects neuronal differentiation. We found that depletion of HDAC4, an N-terminal lysine acetyltransferase, promotes neurite formation in P19 cells, in vivo, and in dissociated cortical neurons. Add-back of the deacetylase domain, but not the lysine acetyltransferase domain, was sufficient for the observed neurite-promoting activity. HDAC4 knockdown also suppressed apoptosis caused by irradiation in cultured cortical neurons. The transcription of BDNF, a marker for neuronal differentiation, was more induced in HDAC4-depleted neurons than in control neurons, whereas BDNF transcription levels in HDAC4-expressing neurons were diminished by treatment with inhibitors of acetyltransferases, including p300.
